Factors Affecting IVIVC

چکیده
The physicochemical properties of the drug substance can assume a primary role in controlling its dissolution from the dosage form. The aqueous solubility of the drug is one of the major factors that determine its dissolution rate. Some studies concluded that the drug solubility data can be used as rough predictor of the possibility of any future problems with bioavailability. Some of the more prominent physicochemical properties of the drug that influence the dissolution rate are discussed below.

Physiologically-Based IVIVC of Azithromycin
Aim of the study is to establish physiologically-based in vitro in vivo correlation (IVIVC) of azithromycin, a biopharmaceutics classification system (BCS) class II drug (high permeability/ low solubility). In vitro dissolution was done using USP apparatus II in pH 6 phosphate buffer at 50 rpm. In vivo pharmacokinetic study was done on 28 healthy humans after IRB and Jordan FDA approvals. Plasm...
متن کاملIn Vitro in Vivo Correlation Basis and Application to Slow Release Injectable Formulation, a Review
The aim of the present paper is to present the basic knowledge about in vitro in vivo correlations (IVIVC). The various definitions are given as well as the requirements to establish IVIVC. The basic concepts and IVIVC use and limitations are described. Application to slow release injectable formulations is presented at the end of the paper as a particular example of IVIVC.
متن کاملIn vitro - in vivo correlation: from theory to applications.
A key goal in pharmaceutical development of dosage forms is a good understanding of the in vitro and in vivo performance of the dosage forms. One of the challenges of biopharmaceutics research is correlating in vitro drug release information of various drug formulations to the in vivo drug profiles (IVIVC). Thus the need for a tool to reliably correlate in vitro and in vivo drug release data ha...
متن کاملModelling and simulation approaches for waiving in vivo pharmacokinetic formulation studies
The bioavailability and bioequivalency of oral drug depends on gastrointestinal tract physiology and drugrelated physicochemical and pharmacokinetic factors. In general, bioavailability of a new drug substance or new formulation is studied in vivo with healthy volunteers. In vivo bioequivalency studies are needed for generic drug products or if a formulation is significantly altered during clin...
